中文摘要:为了筛选出麦二叉蚜Schizaphis graminum参与吡虫啉或氟啶虫胺腈代谢调节的细胞色素P450(P450)基因,采用浸渍法,用吡虫啉或氟啶虫胺腈处理麦二叉蚜24 h后,通过构建转录组测序文库,并进行RNA-seq测序,筛选出差异性表达的P450基因,并进行实时荧光定量PCR(qPCR)验证。结果表明:用吡虫啉处理麦二叉蚜后,有19个P450基因上调表达,15个P450基因下调表达;用氟啶虫胺腈处理麦二叉蚜后,有18个P450基因上调表达,15个P450基因下调表达。运用qPCR验证了吡虫啉对12个P450基因的诱导表达,其结果与DGE测序文库显示的结果基本一致。这些结果可为深入研究麦二叉蚜P450对蚍虫啉和氟啶虫胺腈的解毒代谢作用提供依据。 |

Effects of imidacloprid or sulfoxaflor on the expression of cytochrome P450 genes in greenbug (Schizaphis graminum Rondan) based on RNA sequencing |

Abstract:To identify the cytochrome P450 (P450) genes involved in the metabolic regulation of imidacloprid or sulfoxaflor, the greenbug was treated with imidacloprid or sulfoxaflor for 24 h using the dipping method. Then the differentially expressed P450 genes were identified through transcriptome sequencing and quantitative real-time PCR (qPCR). The results indicated that the expression of 19 P450 genes were up-regulated and 15 P450 genes were down-regulated in the greenbug treated with imidacloprid. In terms of the greenbug treated by sulfoxaflor, the expression of 18 P450 genes were up-regulated and 15 P450 genes were down-regulated. Furthermore, 12 P450 genes affected by imidacloprid were validated by the qPCR method, and the results demonstrated that the expression levels of these genes were consistent with that of the DGE library. These results provide a basis for further study of the detoxification of imidacloprid and sulfoxaflor by P450s in the greenbug. |
